Description: Lipid-based drug delivery systems such as SEDDS are a well-established approach for drug delivery; however, their large design space – (co-)surfactant(s), API type and content, lipid(s), etc. – complicates formulation design. Here, we introduce a digital workflow utilizing Hydrophile-Lipophile Difference/ Net-Average Curvature (HLD-NAC) to predict emulsion phase behavior for rational SEDDS design. We demonstrate the utility of this using in-silico formulation tools to design and experimentally verify a fully-dilutable SEDDS formulation.
